Boxed Warnings: The Most Important Risks

The FDA requires Prolate to carry a black box warning — the most serious warning on a drug label. These warnings highlight risks that can be life-threatening:

Addiction, abuse, and misuse: Oxycodone, the opioid component of Prolate, has high potential for addiction and misuse, which can lead to overdose and death even in patients taking it as prescribed.

Life-threatening respiratory depression: Oxycodone can slow or stop breathing. This risk is highest when starting Prolate or when doses are increased. Risk is even higher if combined with alcohol, benzodiazepines, or other CNS depressants.

Accidental ingestion by children: Even one tablet can be fatal to a child. Store Prolate locked away from children and others.

Hepatotoxicity from acetaminophen: Taking more than 4,000mg of acetaminophen per day — from Prolate or any other source — can cause severe or fatal liver damage.

Common Side Effects (Expected and Manageable)

Most patients taking Prolate will experience at least some of these common side effects, especially when starting the medication or adjusting the dose:

Constipation: The most consistent opioid side effect. Unlike most other opioid side effects, constipation typically does not improve with time. Patients often need a stimulant laxative (e.g., senna) while taking Prolate.

Drowsiness and sedation: Common, especially at the start. Do not drive or operate heavy machinery until you know how Prolate affects you.

Nausea and vomiting: Often improves over the first few days. Taking Prolate with food may help reduce nausea.

Dizziness and lightheadedness: Especially when standing up quickly (orthostatic hypotension). Rise slowly from sitting or lying positions.

Headache

Itching (pruritus): A common opioid side effect caused by histamine release. Usually not an allergic reaction.

Dry mouth

Sweating

Serious Side Effects — Call Your Doctor

Contact your doctor if you experience any of the following:

Shallow or very slow breathing (less than 12 breaths per minute)

Extreme sleepiness or difficulty waking up

Yellowing of the skin or eyes (jaundice) — may indicate liver damage from acetaminophen

Severe abdominal pain (upper right side) — may indicate liver toxicity

Worsening pain with increased doses (possible opioid-induced hyperalgesia)

Confusion, extreme agitation, or rapid heart rate — possible serotonin syndrome if taking serotonergic drugs

Severe skin rash — may indicate a rare but serious skin reaction (Stevens-Johnson syndrome)

Call 911 Immediately If You Notice:

Someone is unresponsive or cannot be woken up after taking Prolate

Breathing has stopped or become gasping

Lips or fingernails are blue (cyanosis)

If you have naloxone (Narcan) available and suspect opioid overdose, administer it immediately and still call 911. Naloxone may need to be repeated as its effects can wear off before oxycodone does.

Watch Your Total Acetaminophen Intake

Each Prolate tablet contains 300mg of acetaminophen. The maximum recommended daily dose of acetaminophen is 4,000mg — and ideally no more than 3,000mg for regular use. If you take Prolate 10mg/300mg four times per day, you're consuming 1,200mg of acetaminophen from Prolate alone. This leaves room for small amounts from other sources, but be careful not to also take Tylenol, NyQuil, or other APAP-containing products without counting toward your daily limit.
